Queensland wicketkeeper Chris Hartley will be ruing the timing of his double batting failure in front of national selector Trevor Hohns in the side’s Sheffield Shield clash with New South Wales at the Gabba. Especially after copping what appeared to be a dubious caught-behind decision that led livid Bulls coach Stuart Law to seek out match referee David Talalla. At stumps on day three, Queensland was reeling at 7 for in their second innings with an overall lead of after Hartley was again sent packing for a duck. Hartley was expected to be in the mix if Test gloveman Brad Haddin did not overcome a shoulder injury in time for the first Test against India starting on December 4. But Hartley appeared to be unfairly denied a chance to impress resident selector Hohns with the bat before the Test squad is named on Monday. He was controversially given out for a nine-ball duck off Doug Bollinger 2 for 15 by umpire Shawn Craig. Hartley left a rising delivery and appeared to be struck on the chest but was given out caught behind by Craig, prompting Law to seek out Talalla in the Gabba stands for clarification. Hartley, who only lasted two balls in the first innings, cut a shattered figure as he trudged off.

The emergence of proteomics, the large-scale analysis of proteins, has been inspired by the realization that the final product of a gene is inherently more complex and closer to function than the gene itself. Shortfalls in the ability of bioinformatics to predict both the existence and function of genes have also illustrated the need for protein analysis. Moreover, only through the study of proteins can posttranslational modifications be determined, which can profoundly affect protein function.

Proteomics has been enabled by the accumulation of both DNA and protein sequence databases, improvements in mass spectrometry, and the development of computer algorithms for database searching. In this review, we describe why proteomics is important, how it is conducted, and how it can be applied to complement other existing technologies. We conclude that currently, the most practical application of proteomics is the analysis of target proteins as opposed to entire proteomes.

This type of proteomics, referred to as functional proteomics, is always driven by a specific biological question. In this way, protein identification and characterization has a meaningful outcome. We discuss some of the advantages of a functional proteomics approach and provide examples of how different methodologies can be utilized to address a wide variety of biological problems. This review is intended to give the molecular biologist a rudimentary understanding of the technologies behind proteomics and their application to address biological questions.

We were faced with the task of trying to understand the significance of these proteins, and the only obvious way to begin to do this was to identify them by sequencing. We then bought an Applied Biosystems automated Edman sequencer not having the budget for a mass spectrometer at the time. Since the majority of intact eukaryotic proteins are not immediately accessible to Edman sequencing due to posttranslational N-terminal modifications, we invented mixed-peptide sequencing This method, described in detail later, essentially enables internal peptide sequence information to be derived from proteins electroblotted onto hydrophobic membranes.
